What is the easiest way to remove the front door window regulator and glass.
I thought my B body was tough but stripping the Challenger door is a right pain in the butt.

agreed. this task is a challenge and a nightmare.  paying a garage mechanic at $150/hour could take the better part of a day.  So, rest assured, it's worth the effort.  I recall my forearms getting cut up by the sheet metal edges in the door panel while reaching in there to remove the works.

to R&R (remove and replace) the assembly, and if my memory still serves, one procedure I've used is:

1. remove the glass. disconnect the fasteners to remove the glass from the regulator, track and door.

2. remove the regulator and track. disconnect and remove the window regulator and track assemblies.

To install, go in reverse.  try not to break anything.  some of those parts are hard to find.

Following this procedure, I seem to recall it can take (me) a better part of a Saturday to swap a door and move the glass and window regulator parts over.  My forearms will have cuts and bleeding from reaching through them openings in the door.

If you can do this in less time and injury, consider yourself lucky.

Glass out through the top of the door. Regulator out through the big opening in the bottom of inner door sheet metal. Tracks off regulator.

It's been about 10 years so memory is foggy. I do remember though using blocks of wood to hold up the glass and I think I did that so I could wiggle either the regulators or the track out of the way to make more room to extricate the glass. I think everything has to be undone or the glass isn't coming out. It has to come out at an angle too, I believe out the rear of the door. A roller or stud comes out with the window and it has to slip out a wide opening that is only at the back of the door.

I'm going to suggest that you review a really good thread for door glass here on this site. I will see if I can find the link for you, but I used the instructions a number of times, and they're rock solid. I did mine three times, and each time I got better and faster. Also, the shop manual is an excellent resource as well.

If you're going to actually replace the glass, I might suggest ECS glass - it's very expensive, but I am pretty sure it's better than AMD glass. Also, from my experience, the left side AMD glass has the correct curvature in it, but the right side glass does not, and it took me forever to adjust it so it was tracking correctly and also not squeaking or shuddering. I have checked with a few other knowledgeable folks who work on a lot of e-bodies, and they reported the same thing. Good luck w/you door glass.
